The Investigation of MiR-125a and MiR-155 Levels in White Coat Hypertension and Essential Hypertension

Objective:

White coat hypertension (WCH) is a high cardiovascular risk condition and a fundamental understanding of the cause and pathophysiology of the disorder is still lacking. Recent studies demonstrated that microRNAs (miRNAs) are involved in hypertension. However, the roles of miRNAs in WCH are not known. The first aim of this study was to investigate the levels of miR-125a and miR- 155 plasma expression levels in the WCH, hypertension (HT) and normotensive(NT) groups and compare the levels between the groups. The second aim was to evaluate the relationships between these miRNA levels and the measurements of clinical and ambulatory blood pressures.

Material and Methods:

The expressions of miR-125a and miR-155 were validated independently in plasma samples from 30 HT patients, 30 WCH and 30 NT healty subjects. The expression levels were analyzed by quantitative reverse transcription-PCR.

Results:

There was not found any significant difference in terms of expression levels of miR-125a and miR-155 between all the groups. miR-125a levels were significantly positively correlated with miR-155 levels in all groups (r=0,861, p<0,001). No significant differences were observed between the levels of miR-125a and laboratory findings, clinical and ambulatory blood pressure measurements. The levels of miR-155 were only significantly negatively correlated with the measurements of waist circumference.

Conclusion:

Our results were correlated with the literature, however we could not determine statistically significant relationships because of the small number of groups. It was observed that the miRNAs expression levels were positively correlated between HT and WCH patient. These results suggest that the miRNAs might have important role in the epigenetic aspect of ethiopathogenesis of HT and WCH for our understanding.
